Our Mission
To conduct a comprehensive assessment of a population’s current immune response to SARS-CoV-2 through a volunteer-based medical study and publish the findings of our study to the public using an easy to understand and accessible online platform.
Volunteer Blood Assay
Sample 1: HRP tagged SARS-CoV-2 protein added AFTER incubation.
We will collect OD 450 and then with this number calculate the % inhibition. High inhibition % means higher concentration of Neutralizing antibodies (Nab) and a lower OD 450. This is repeated twice, we will provide both OD, but then only one % inhibition.
Sample 2: HRP tagged SARS-CoV-2 protein added PRIOR to 16-24 hours of 37 C incubation.
Volunteers get a results report with 2 valuesof OD 450 and one value of % inhibition for each
sample.
We will also provide the pre/post % inhibitions and the difference between them. We will provide low, medium, high rang values representing 30-60% low inhibition- 60-90% medium inhibition, and above 90% is high inhibition.
Our blood assessment utilizes an enzyme-linked immunosorbent assay (ELISA) to quantitative biochemical assay capable of monitoring the protein-protein interactions of SARS-CoV-2 Spike proteins with ACE-2 host proteins. A standard curve serial dilution of SARS-CoV-2 neutralizing antibodies (Nab) of known concentration is prepared along with both positive and negative controls. The ELISA plates are coated with ACE-2 proteins, which will bind with the receptor binding domain (RBD) portion of the SARS-CoV-2. The SARS-CoV-2 proteins in this assessment are coated with horseradish peroxidase (HRP). The HRP will generate colored compounds when
it is exposed to light with a 450nm filter. The concentration of the colored compounds are
measured, the optical density (OD) of the sample is compared to the standard curve.
The percent of inhibition from Nab are calculated:
% Inhibition =(1- OD value of sample / OD value of Background) x 100%
